ABSTRACT

Objective To observe in vitro the effect of mechanical stress at different intensities on the pro-liferation, apoptosis and extracellular matrix of degenerative human nucleus pulposus cells. Methods The cells were isolated and cultured in vitro, and divided into a control group, a low-intensity group, a medium-intensity group and a high-intensity group. The low-, medium- and high-intensity groups were stretched mechanically by 1000 μ, 2000μor 4000μrespectively for 6 hours using a four-point bending system, while the control group was not stressed. Flow cytometry was used to explore any changes in the cell cycle and the proliferation index ( PI) . The expression of proliferating cell nuclear antigen ( PCNA) , B-cell lymphoma-2 ( BCL-2)/Bax, collagen II and aggrecan were meas-ured using real-time quantitative polymerase chain reactions. Results The mechanical stretching significantly influ-enced proliferation, apoptosis and the extracellular matrices compared with the control group. The PI and PCNA ex-pression increased at first and then decreased gradually with the exercise intensity. Compared with the control group, the mRNA expression level of Bcl-2/Bax increased significantly to 1.53 times that of the control group after 1000 μstretching, but to only 0.71 times that of the control group at 2000μand 0.43 times at 4000μ. The gene expression of collagen II increased significantly by 1. 1 times and that of aggrecan by 1. 3 times after 1000 μ stress stimulation compared with the control group ( P≤0.05) . However, the expression of collagen II and aggrecan was inhibited sig-nificantly at 2000 and 4000 μ , with the lowest levels at 4000 μ (P≤0.05). Conclusion Stretching at different intensities has different effects on the proliferation, apoptosis and extracellular matrix of human pulposus cells with degenerate nuclei.

ABSTRACT

Objective To investigate the treatment and mechanism of aminoguanidine in retina of diabetic of rats .Methods To-tal 60 rats were divided into control group(n=20) ,diabetic group(n=20) and aminoguanidine treatment group(n=20)which would be treated by aminoguanidine for 14 days .Then the eye tissue of rats were took after 14 days administration for pathological obser-vation(HE staining) ,and the induced nitric oxide synthase(iNOS) ,endothelial nitric oxide synthase(eNOS) ,nerve type of nitric ox-ide synthase(nNOS) level and the expression of differences content and expression were investigated by ELISA ,Western blot and PT-PCR .Results HE staining showed that retinal tissue defects decreased and neuronal cells of rats in aminoguanidine treatment group were increased and significant (P 0 .05) .Compared with diabetes group ,iNOS ,eNOS ,nNOS protein expression in the rat retina in aminoguanidine treatment group were reduced (P0 .05) .The iNOS mRNA expres-sion was lower than that of eNOS mRNA and nNOS mRNA in aminoguanidine treatment group .Conclusion Aminoguanidine can improve retinal tissue of diabetic rats with lesions ,the pathways may be selectively inhibit inducible nitric oxide synthase activity of iNOS .

ABSTRACT

Purpose of this study was to validate that Subtenon [SB] Triamcinolone [TA] injection is an alternative to Intravitreal [IV] Triamcinolone [TA] injection for the treatment of diabetic macular edema [DME]. Forty eyes were selected having DME due to type 1 or type 2 diabetes. All the patients were treated with photocoagulation. IVTA was administered in one eye and SBTA in following eye of same patient. Improvement in visual acuity, macular edema and intraocular pressure was assessed before treatment and on 2[nd], 4[th], 8[th] and 12[th] week after treatment. After administration of IVTA, MVA was reduced from baseline value [0.805 +/- 0.069Log/MAR] to [0.577 +/- 0.091 Log/MAR, p < 0.001] at the end of treatment. Similar results were observed after SBTA administration. MVA was reduced from [0.814 +/- 0.082Log/MAR] to [0.49 +/- 0.080 Log/MAR, p < 0.001] at 12th week. After IVTA injection Central macular thickness was significantly reduced to [246.8 +/- 25 microm, p < 0.001] from [390.5 +/- 17 microm]. There were no significant [p=0.51] difference in both eyes receiving different routes of same treatment. After SBTA injection CMT was significantly reduced to lower values [241.5 +/- 27 microm, p < 0.001] from [394.4 +/- 21 microm]. Intraocular pressure after IVTA administration was high [2.32 +/- 0.72 mm/Hg, p=0.04] as compared to baseline [1.82 +/- 0.94 mm/Hg]. Similar pattern was also seen after SBTA administration but to significant extent. Elevation of IoP was observed in both eyes.
